Intracellular Distribution of Nonstructural Protein 2 in the Spinal Cords of Mice Infected with Neuroadapted Sindbis Virus

Abstract
Purpose: The Neuroadapted Sindbis virus (NSV) induces an acute viral encephalomyelitis in susceptible mice. Nonstructural protein 2 (nsP2) of the Neuroadapted Sindbis virus (NSV) has been suggested to play an essential role in virus-host cellular interactions and for the formation of individual nonstructural proteins. Until present, the intracellular localization of NSV nsP2 has been studied with cultured cells. This study aims to examine the intracellular localization of NSV nsP2 in the spinal cords of the mice after infection.

Materials and Methods: Five to seven-week old C57BL6/J mice were infected intra-nasally with NSV. At 3, 5, and 7 days after infection, the cervical and lumbar spinal cords were taken out after perfusion fixation. Paraffin sections were prepared and immuno-stained for nsP2.

Results: NsP2-immunoreactivities (ir) were observed exclusively in the nuclei of infected neurons of the cervical and lumbar spinal cords 5 days after infection. These nsP2-labeled nuclei showed a healthy-looking euchromatic appearance. At a later time point, day 7 of post-infection, it was still localized in the neuronal nuclei of the lumbar cord while being observed in the cytoplasm of the cervical cord. Cells with cytoplasmic nsP2 showed condensed features.

Conclusion: The intracellular distribution of NSV nsP2 in the spinal cord is different between early and later time points after infection. The different localization patterns between the cervical and lumbar cords at the later time point is related to the activity of the viral replication, which may lead to different progress of the neuronal degeneration.
